The breakthrough came from combining three advances: mRNA technology, checkpoint inhibitor drugs, and affordable genetic sequencing. Previous cancer vaccine attempts failed because they only targeted one or two tumor mutations, but this vaccine targets 34 personalized mutations in each patient.
The vaccine works by carrying genetic instructions that direct the immune system toward a patient's specific cancer cell mutations. Doctors create it using a sample of each person's tumor, making every dose unique.
Melanoma was chosen for the trial because this skin cancer has many mutations and already responds well to immunotherapy drugs like Keytruda. Patients received the vaccine after surgery, giving their immune systems time to mount a defense before cancer could spread.
