
Yale Scientists Find Way to Fight Aging Without Dieting

A new study in Nature Aging points to a promising answer. Yale researchers studied 42 people who cut their calorie intake by just 11 to 14% for two years as part of a rigorous NIH-funded trial. The participants didn't feel deprived and stayed healthy.
The team measured over 7,000 proteins in blood samples collected throughout the study. One protein stood out: complement component 3, or C3. This immune protein dropped significantly after moderate calorie restriction.
C3 caught the researchers' attention because it contributes to chronic inflammation, which drives many age-related diseases. But no one had proven C3 actually caused aging problems until now.

The team discovered something unexpected about where C3 comes from. White fat tissue, especially around the organs, was the main source. Even more surprising, the C3 was being made by specific immune cells called macrophages that live in fat tissue.
Here's where it gets really exciting. The researchers initially thought losing weight might be what reduced C3 and improved health. Most study participants lost about 18 pounds over two years.
But when they compared weight loss with C3 reduction, they found no connection. The calorie restriction benefits appeared to work independently of dropping pounds.
To test if they could skip the dieting part entirely, the researchers used a drug to block C3 in mice. The animals developed less age-related inflammation, just like mice on calorie-restricted diets.
"This concept demonstrates that aging is actually malleable and a process that can be targeted," says Dr. Vishwa Deep Dixit, who directs Yale's Center for Research on Aging.

The discovery opens the door to future treatments that could help people age more healthily without requiring lifelong calorie restriction.
