Novartis Finds Single-Dose Cure for Chagas in Mice

Six million people live with Chagas disease, a life-threatening parasitic infection, yet fewer than 1% receive proper treatment because current therapies require weeks of medication that often cause unbearable side effects. Now, researchers at Novartis have identified a compound that achieved complete cure in mice after a single oral dose.

The experimental drug, called IID432, works by targeting a specific weakness in the parasite that causes Chagas disease. Unlike existing treatments, it selectively attacks the parasite's DNA management system while leaving human cells completely unharmed.

In laboratory studies published in a major scientific journal, the compound achieved sterile cure in mice with chronic infection. The parasites were eliminated so thoroughly that they never returned, even after researchers monitored the animals long term.

"Millions of people are affected by Chagas disease, yet current treatments for this life-threatening infection can require weeks of therapy, which is often abandoned due to side effects," says Fiona H. Marshall, President of Biomedical Research at Novartis. Her team's discovery opens the possibility of dramatically shorter treatment regimens.

Chagas disease kills 12,000 people annually and is endemic across 21 Latin American countries. The infection often goes unrecognized during its early stages, then progresses silently for years before attacking the heart and digestive system.

Current medications can take 60 days to complete and frequently cause side effects severe enough that patients stop treatment. This new approach could transform a two-month ordeal into a single doctor's visit.

The Ripple Effect

Beyond the laboratory, Novartis has been strengthening healthcare systems across Latin America to better diagnose and treat Chagas disease. Since 2019, the company has partnered with Brazilian researchers on a major study involving people with heart failure caused by chronic Chagas infection.

The company is also working with global health organizations including the World Heart Federation and Global Chagas Disease Coalition to build medical capability in affected communities. These partnerships ensure that when new treatments become available, healthcare systems will be ready to deliver them.

The research team designed IID432 to form a precise chemical bond with a protein found only in the parasite, not in humans. This selectivity is what makes the compound both highly effective and potentially safe.

While additional research is needed before human trials can begin, the findings provide strong evidence that dramatically shorter Chagas treatment is scientifically possible. The work was supported by Novartis Biomedical Research and the Wellcome Trust.
